Spectroscopy and coherent control in biological systems

We develop and apply spectroscopic techniques based on femtosecond lasers in order to shed light on structural dynamics of proteins during biochemical processes. The association of mid-infrared femtosecond pulses with state-of-the-art femtosecond technologies such as pulse shaping and multidimensional spectroscopy offer a powerful tool to investigate biomolecules such as hemoproteins.

Currently ongoing projects include:

- Ultrafast mid-infrared pump-probe spectroscopy, which allows to investigate with a femtosecond resolution structural changes acommpanying ligand transfer in biomolecules such as hemoproteins.  
 
 
Two-Dimensional Infrared Spectroscopy (2DIR), a transposition of 2D NMR to nonlinear optical spectroscopy, is sensitive to protein structure fluctuations through the resulting fluctuation of the oscillation frequency of vibrational local probes, such as carbon monoxide in hemoproteins. We have also demonstrated the application of transient 2DIR in the context of vibrational ladder climbing.
 
Vibrational Ladder Climbing consists of the excitation of an anharmonic vibrational ladder with a chirped mid-infrared pulse, allowing to drive a molecular vibration far from equilibrium. We have demonstrated the excitation of carboxy-hemoglobin up to the seventh vibrational level.
 
-L’AD-ASOPS (Arbitrary Detuning-ASynchronous Optical Sampling) is a new pump-probe method (patent pending) based on the use of two independent femtosecond sources, either two free-running femtosecond oscillators, or two femtosecond amplifiers seeded by independent and free-running oscillators. It allows to measured the dynamic response on timescales ranging from sub-picosecond up to millisecond.
